Regionals will continue to flourish within the permitted scope as long as they are able to staff the flying, remain cost competitive and there are no other constraints like infrastructure congestion at large airports.
The fact that mainline upgauges a route just means that customer demand / preferences justifies an equipment change. Maybe there is more traffic, maybe more business / first class revenue or simply a competitive response because someone else is flying an A320 on that route now.
The newly freed up regional equipment will just re-deployed somewhere else. There are plenty of places today that could support some EAS or other service. So the plane that just got replaced by mainline will be utilized elsewhere as long as marginal revenue exceeds marginal cost.
Mainline seniority lists may grow but not at the expense of regional seniority lists – again, this assumes that there are no other constraining factors.
